Semaglutide targets one receptor, tirzepatide two and retatrutide three. That structural distinction is where every other difference originates.

Regulatory status differs materially
Semaglutide and tirzepatide hold marketing authorisation in the European Union and are prescribed there as medicines. Retatrutide does not: it is in clinical development and is not authorised as a medicine anywhere.
