NAD+ (nicotinamide adenine dinucleotide) is strictly speaking not a peptide but a coenzyme: a small molecule present in nearly every cell and indispensable to energy metabolism. It shuttles electrons between reactions, alternating between the NAD+ and NADH forms.
Beyond that metabolic role, NAD+ is the substrate for two enzyme families that attract considerable attention in ageing research: the sirtuins and the PARPs, involved in gene regulation and DNA repair respectively. Both consume NAD+, which is why its availability is studied as a limiting factor.
Measured NAD+ concentration in tissue declines with age. That observation is what the research field is built on; what follows from it is the subject of ongoing work, not settled fact.
